Broad spectrum plasmid transfection

TransIT®-LT1 Transfection Reagent

A broad spectrum, low toxicity, DNA transfection reagent

The TransIT®-LT1 (Low Toxicity) Transfection Reagent is Mirus Bio’s most popular broad spectrum plasmid delivery reagent, proven to achieve high transfection efficiencies in a wide variety of cell types with minimal cellular toxicity. The reagent is a proprietary formulation of histone and lipid compounds. Highest transfection efficiencies are achieved when transfections are performed in serum-containing media, with no media change. This robust reagent is supplied with an optimized protocol as a ready-to-use single reagent, which allows for ease of use and high reproducibility. MIR 2300 provides sufficient reagent to perform up to 500 transfections in 35 mm dishes or -well plates.

TransIT®-2020 Transfection Reagent

A high performance, animal-free, broad spectrum DNA transfection reagent

TransIT®-2020 is a broad spectrum transfection reagent that provides superior transfection of plasmid DNA into mammalian cells. TransIT-2020 is suitable for both transient and stable transfection and works well in typically hard-to-transfect cell lines. TransIT-2020 is comprised of animal-origin free components and is serum compatible, eliminating the need for any culture media change after transfection.

Experimental Details: Luciferase gene expression was compared in HEK 293, K562, CHO-K1, and primary MEF cells transfected with a luciferase expression plasmid using TransIT®-2020 (Mirus Bio, 3:1 reagent to DNA ratio), FuGENE® HD (Roche, 7:2 reagent to DNA ratio), Lipofectamine™ 2000 (Invitrogen, 5:2 reagent to DNA ratio), and Lipofectamine™ 2000 CD (Invitrogen, 5:2 reagent to DNA ratio). Transfections were performed in 24-well plates using 0.5 µg of plasmid DNA per well. The optimal level of each transfection reagent was determined empirically and all reagents were used according to manufacturer’s protocol. Cells were harvested at 24 hours post-transfection and assayed for luciferase activity. Error bars represent the standard deviation of triplicate wells.
