Mirus Bio Protein Production & Expression Systems

High-Yield Transient Protein Expression Systems for Suspension Cells
Mirus Bio Cell culture & transfection

The Mirus Bio CHOgro® High Yield Expression System is an optimized platform specifically engineered to enhance transient protein expression in suspension CHO cell cultures. By combining high-density cell growth with a high-efficiency transfection protocol and a specialized titer enhancer, this system enables researchers to achieve significantly higher protein yields (up to 10-fold) compared to standard methods. The entire workflow is animal-origin-free and designed for quick implementation without the need for complex licensing agreements.

Technical Guidelines

Operational Considerations & Optimization

CHOgro Expression Medium requires supplementation with 4 mM L-Glutamine and 0.3% Poloxamer 188 prior to use. Suspension CHO lines typically adapt directly or via step-wise sequential adaptation (e.g., 75/25 to 100% ratios). Passaging should be performed to maintain cell densities between 2.5 x 10^5 and 1 x 10^7 cells/ml.

For optimal results, passaging 18-24 hours prior to transfection ensures active cell division. Target seeding density is 4 x 10^6 cells/ml at the time of transfection. Complex formation should occur in CHOgro Complex Formation Solution with a 5-minute incubation period at room temperature. Post-transfection, an immediate shift to mild hypothermal conditions (32 degrees C) enhances specific productivity and extends culture longevity.

Frequently Asked Questions

Key technical and operational details regarding vector production reagents and scaling.

Yes, TransIT-VirusGEN® reagents and kits are formulated for high-yield transfection in both adherent, serum-containing cultures and serum-free suspension HEK 293 systems. Dedicated complex formation guidelines are provided for each culture format

RevIT™ AAV Enhancer works synergistically with TransIT-VirusGEN® transfection reagents to increase AAV vector yields 2- to 4-fold in suspension HEK 293 workflows. It can be added during complex formation or introduced directly into the culture.

TransIT-VirusGEN® GMP reagents and kits undergo quality control testing for appearance, formulation identity, sterility, bacterial endotoxins, mycoplasma, pH, and osmolality to support clinical manufacturing requirements.

No media change is necessary post-transfection. Transfection complexes remain in the culture medium throughout the full incubation period without compromising cell health or viral titers.

A complex formation incubation of 15–30 minutes at room temperature serves as a standard starting point. For large-scale vessel processing, complex formation times can be evaluated between 5 and 60 minutes based on specific construct dynamics.

TransIT-VirusGEN® reagents and RevIT™ Enhancers must be stored frozen at -10°C to -30°C. Complex formation solutions and LV enhancers are stored at 2°C to 10°C. Reagents should be warmed to room temperature and mixed gently before complex preparation.
