The seqWell MosaiX™ 90-minute workflow represents a significant leap in NGS library preparation by bridging the gap between the speed of tagmentation and the high-quality data typically reserved for ligation-based methods. At its core is the engineered TnX transposase, which has been evolved to drastically reduce the sequence-dependent insertion bias common in traditional Tn5 methods.
The process is remarkably efficient, requiring only 35 minutes of hands-on time. It utilizes a proprietary directional tagmentation chemistry that ensures every tagging event produces a functional, sequencer-ready molecule (with the proper P5 and P7 orientations), effectively doubling the molecular complexity of the library compared to older tagmentation kits. The kit is highly robust and compatible with DNA diluted in 10 mM Tris-HCl, 1X TE, Low TE, or Water. Its fragmentation profiles remain unaffected by EDTA.
