Guidance on selecting the optimal library prep chemistry for your sample type, input requirements, and workflow goals.

Selection depends on your research goals and sample quality. Choose mRNA Library Prep for focused coding-region analysis of high-quality (RIN >7) eukaryotic samples. Select RNA Library Prep (Total) for targeted sequencing or hybridization capture from ultra-low inputs (0.25 ng). Use RNA Prep with Polaris Depletion for whole-transcriptome sequencing from blood or degraded samples where removal of rRNA and globin transcripts is required.

Watchmaker kits include a specialized FFPE Treatment Buffer designed to address chemical damage and cross-linking inherent in fixation. When paired with the engineered StellarScript™ Reverse Transcriptase, this module enhances conversion efficiency, allowing for the generation of high-complexity libraries from samples that typically fail with standard RT enzymes.

The Polaris Depletion module is specifically optimized for Human, Mouse, and Rat (HMR) samples. It efficiently removes cytoplasmic and mitochondrial rRNA across all three species, while its globin-specific probes are specifically designed for Human transcripts.

Yes. The workflows are designed with fewer bead cleanups and "on-magnet" washing steps to minimize handling. Watchmaker provides 96-reaction kits with generous overages to accommodate dead volumes on liquid handling platforms like the Biomek i7.

These kits utilize random priming for first-strand synthesis to ensure even coverage across the transcript, which typically produces library inserts of ~200 bp. Because they do not generate full-length cDNA, they are not recommended for native long-read sequencing applications.
